Info: Tirzepatide is a synthetic peptide and dual agonist of the GIP and GLP-1 receptors. It is designed to mimic the activity of naturally occurring metabolic hormones involved in glucose regulation, insulin secretion, appetite control, and energy balance. Tirzepatide consists of a 39-amino-acid peptide sequence modified through lipidation, which contributes to its prolonged activity and metabolic stability. 

Effects

Glucose Regulation: Tirzepatide activates both GIP and GLP-1 receptors, influencing glucose-dependent insulin secretion and glucagon regulation. This dual mechanism supports research into blood-glucose control and metabolic function.

Side Effects

Common side effects of Tirzepatide may include gastrointestinal symptoms such as nausea, vomiting, diarrhea, constipation, abdominal discomfort, bloating, and indigestion, particularly during dose escalation.

Dosage Range and Duration of Use

Tirzepatide is administered by subcutaneous injection, generally once weekly. The usual clinical starting dose for adults is 2.5 mg once weekly for the first 4 weeks, followed by 5 mg once weekly. Depending on therapeutic response and tolerability, the dose may subsequently be increased in 2.5 mg increments at intervals of at least 4 weeks, with maintenance doses of 5 mg, 10 mg, or 15 mg according to the approved indication. The maximum recommended dose is 15 mg once weekly.

Treatment duration varies according to the approved indication, individual response, tolerability, and clinical requirements. 

Combination with Other Drugs

Tirzepatide may interact with other medications, particularly insulin and certain glucose-lowering medicines, potentially increasing the risk of hypoglycemia. It may also affect the absorption of orally administered medicines by delaying gastric emptying.

Tirzepatide is contraindicated in patients with a personal or family history of medullary thyroid carcinoma or in individuals with Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). It is not indicated for the treatment of type 1 diabetes.
